Methodology

Bone marrow or peripheral blood samples with circulating blasts cells are set up in 24 and 48 hour unstimulated cultures, unless a B cell malignancy is suspected. For suspected B cell malignancies, samples are set up in 24 hour unstimulated cultures, 72 hour TPA and 120 hour IL4 B cell mitogen stimulated cultures.

Solid tumor: Direct suspension cultures and in situ fibroblast cultures from collagenase treated disaggregated cells are established.

Following the arrest of the cell division with Colcemid, chromosomes are stained by the G-banding method. The chromosome number is determined by microscopic analysis and these cells are examined for the presence or absence of detectable structural rearrangements. Results include banded karotypes for each abnormal clone and the normal cell line if present and analysis of twenty or more metaphases.
